Filter Requirements for Confocal Microscopy
Confocal microscopy puts unusual demands on optical filters, where each filter type in the light path has its own job to do.
Excitation filters clean up the laser line before it reaches the specimen, typically as a narrow ~10 nm bandpass.
Dichroic mirrors need high flatness to keep the beam quality intact under high laser power.
Emission filters carry the heaviest load, often blocking the excitation wavelength at OD8 or higher while still passing the fluorescence signal.
Chroma Technology Corp. assembled a breakdown of all three of these filter types in their “Confocal Microscopy Application Guide.”
